Revamped Remedy for Tough-to-Tame High Blood Pressure Shows Promise

The global clinical trial Phase 3 Launch-HTN has made waves in the medical world with its findings, revealing that lorundrostat effectively and safely controls blood pressure in a wide range of patients who previously resisted other treatments.

High blood pressure, or hypertension, poses a significant risk to public health by increasing the likelihood of heart disease, heart attacks, and strokes. This condition is particularly troublesome when it becomes resistant to common medications, unfortunately affecting up to 15% of individuals with hypertension.

In response to this issue, researchers have developed lorundrostat - a drug that targets and inhibits aldosterone synthase, the enzyme responsible for producing aldosterone, a hormone involved in regulating blood pressure. The study showed consistent reductions in blood pressure across a diverse group of participants, marking the largest phase three trial within this treatment class.

Dr. Manish Saxena, Clinical Co-Director of the William Harvey Heart Centre at Queen Mary University of London and Lead Investigator of the study, had this to say during an interview with Medical News Today:

"With an abundance of treatments available, more than half of adults with hypertension worldwide remain unable to reach their blood pressure goals. Dysregulation in the aldosterone pathway plays a crucial role in blood pressure regulation and leads to complications such as heart failure and kidney problems. The Launch-HTN trial aimed to explore the safety and efficacy of lorundrostat, a novel drug that belongs to a new class of aldosterone synthase inhibitors."

Lorundrostat consistently lowered blood pressure in study patients, dropping systolic blood pressure by 16.9 mmHg at Week 6 and by 19 mmHg at Week 12. These reductions correspond to a 9.1 mmHg decrease compared to placebo at Week 6 and an 11.7 mmHg decrease versus placebo at Week 12.

"The Launch-HTN trial represents a significant stride in our fight against hypertension, as it tests lorundrostat in a diverse patient population and demonstrates its favorable safety profile and consistent efficacy," Saxena explained.

Other researchers, not associated with the trial, also shared their perspectives on the study with MNT:

  • Cheng-Han Chen, MD - board-certified interventional cardiologist and medical director of the Structural Heart Program at MemorialCare Saddleback Medical Center in Laguna Hills, CA - emphasized that this trial signifies one more tool in the ongoing battle against stubborn blood pressure. "For patients who struggle with uncontrolled or resistant hypertension despite multiple medications, having another class of blood pressure medication in our therapy arsenal will help us better manage this condition and improve long-term health outcomes," she said.
  • Rigved Tadwalkar, MD, FACC - consultative cardiologist and director of Digital Transformation at Pacific Heart Institute in Santa Monica, CA - highlighted that lorundrostat's mechanism-specific approach could change the hypertension treatment landscape. "By inhibiting aldosterone synthesis at the enzymatic level, lorundrostat shows promise for patients who have failed to find relief through standard therapy. The significant reductions in blood pressure, particularly for those with resistant hypertension, signal a meaningful advancement in hypertension treatment," Tadwalkar concluded.

Saxena expresses optimism about the future of lorundrostat, stating, "When lorundrostat becomes commercially available, it has the potential to provide a novel treatment option for patients with hypertension, offering new hope for those who have exhausted other treatment pathways."
